PTI Introduces IntelliSynth UV-Monitoring and Feedback Control System

Protein Technologies, Inc. (PTI) is revealing its breakthrough technology for the Tribute peptide synthesizer at the American Peptide Symposium in Bloomington, IN.

According to PTI, the new IntelliSynth UV-Monitoring and Feedback Control System is a powerful new technology that can monitor the efficiency of Fmoc removal during deprotection reactions by measuring the concentration of dibenzofulvene adduct formed.

Users can view and print monitoring data in real-time, and store it for later use. Data from individual deprotection reactions can be viewed, as well as an overall efficiency summary for the synthesis. Two feedback control options automatically modify deprotection and/or coupling cycles based on feedback. This minimizes synthesis times and solvent usage, as well as trial-and-error especially when synthesizing difficult peptides.

When paired with the PTI peptide predictor software (which predicts the difficulty of coupling reactions based on the peptide sequence), Tribute users will be able to identify difficult cycles before, during, and after a synthesis.

PTI’s UV-monitoring system is the first designed for batch synthesizers rather than continuous flow systems. This offers several advantages over previous technologies: It can be placed on synthesizers with more than one reaction vessel, and the measurement area is automatically rinsed when the resin is rinsed.

In addition, with PTI’s system, it is not necessary to calibrate nitrogen or solution flow rates, making it user friendly. Existing instruments can be retro-fitted in the field, and a simple software upgrade gets users up and running, making the IntelliSynth System a solution for new and existing Tribute users.
